Detection of Sperm DNA Damage in Workers Exposed to Benzene by Modified Single Cell Gel Electrophoresis

چکیده
Objective To assess the effect of benzene on sperm DNA damage Methods Twenty-seven benzene-exposed workers were selected as exposed group and 35 normal sperm donors as control group. Air concentration of benzene series in workshop was determined by gas chromatography. As an internal exposure dose of benzene, the concentration of trans, trans-muconic acid (ttMA) was determined by high performance liquid chromatography. DNA was detected by modified single cell gel electrophoresis (SCGE). Results The air concentrations of benzene, toluene and xylene at the workplace were 86.49 ± 2.83 mg/m3, 97.20 ± 3.52 mg/m3 and 97.45 ± 2.10 mg/m3, respectively. Urinary ttMA in exposed group (1.040 ± 0.617 mg/L) was significantly higher than that of control group (0.819 ± 0.157 mg/L). The percentage of head DNA, determined by modified SCGE method, significantly decreased in the exposed group (n=13, 70.18% ± 7.36%) compared with the control (n=16, 90.62% ± 2.94%)( P<0.001). Conclusion The modified SCGE method can be used to investigate the damage of sperm DNA. As genotoxin and reprotoxins, benzene had direct effect on the germ cells during the spermatogenesiss.

Assessment of DNA damage induced by butachlor on Cyprinuscarpio(L. 1758) using Single Cell Gel Electrophoresis
Butachlor (N-(butoxymethyl)-2-chloro-N-2, 6-diethylacetanillide) is widely used chloroacetanilide herbicide in Asian region, especially in the north of Iran. Several studies have been reported on toxicity of butachlor, but little information is accessible on DNA damaging potential. Thus, the present study was undertaken to investigate of DNA damage by butachlor in Cyprinuscarpio(Pisces: Cyprini...
متن کاملEvaluation of DNA damage in Cyprinus carpio (L. 1758) exposedto malathion using Single Cell Gel Electrophoresis
Malathion (S-(1, 2-dicarboethoxyethyl) O, O-dimethyl phosphorodithioate) is one of the organophosphate pesticides comprehensively used in agriculture fields throughout the world. In spite of widely used, little information are available about DNA damage in aquatic organisms. Therefore, the present study carried out to investigation of DNA damage induced by malathion in Cyprinus carpio (Pisces: ...
متن کاملRadioadaptive response in peripheral blood leukocytes of occupationally exposed medical staff with investigation of DNA damage by the use of neutral comet assay
Background: ˝Radioadaptive Response˝ is well-documented phenomenon appeared in low dose ionizing radiation received in vitro and in vivo. Occupational exposure has always been a great concern for radiation workers therefore this study was performed to study radioadaptive response in terms of residual DNA double strand breaks as an endpoint in peripheral blood leukocytes of occupationally expose...
متن کاملReduced DNA damage in tumor spheroids compared to monolayer cultures exposed to ionizing radiation
Background: Several cell lines when cultured under proper condition can form three dimensional structures called multicellular tumor spheroids. Tumor spheroids are valuable in vitro models for studying physical and biological behavior of real tumors. A number of previous studies using a variety of techniques have shown no relationship between radiosensitivity and DNA strand breaks in monolayer ...
متن کاملNo increased DNA damage in peripheral lymphocytes of sewage workers as evaluated by alkaline single cell gel electrophoresis.
OBJECTIVES To study whether sewage workers are exposed to genotoxic substances. An increased risk of cancers among sewage workers has been noted. If this increased risk is due to an exposure to genotoxic agents, primarily DNA damage could be used as a biological marker of exposure. METHODS In a cross sectional study, DNA damage in peripheral lymphocytes from 35 sewage workers and 30 controls ...
